That i need backer board on top of the pywood, etc. i have read so many opinion that contradict one another. so i lean on the people who know best you guys, single vessel bowl vanity going in a basement bathroom low usuage, how do you suggest build my counter top, plywood and seal it or the tile mortar and grout will do that for me?

If you are going to tile a countertop  in a "wet" area, use 3/4 plywood or MDF, then attach "cement board" and over that apply a coat or two of Kerdi "red guard" or a similar water-proofing agent.

Then tile and grout.

Tile and grout - by themselves - do not waterproof a surface.

I'd avoid MDF completely in a wet area like a bathroom. Particle board as well. The Hardibacker 1/4" backer board on top of exterior grade plywood would be ideal; it holds up extremely well against water.

The ready-to-mix thinset would perform better than the premixed thinset. Small area like the countertop, especially with low usage, the premixed thinset would do okay.
